Modeling Interactions in Circadian Neurons and Peripheral Clocks

چکیده
The recognition of the complexity underlying biological phenomena has renewed interest in systems biology approaches. Due to the critical role biological clocks play in a number of physiologically and clinically relevant situations, we utilize integrative quantitative approaches to reverse-engineer the mechanisms underlying the generation of the circadian rhythms from the synchronization of noisy individual pacemaker neurons and the subsequent oscillatory interaction between the circadian and metabolic transcription networks.

Circadian Gene Expression in Individual Fibroblasts Cell-Autonomous and Self-Sustained Oscillators Pass Time to Daughter Cells
The mammalian circadian timing system is composed of a central pacemaker in the suprachiasmatic nucleus (SCN) of the brain and subsidiary oscillators in most peripheral cell types. While oscillators in SCN neurons are known to function in a self-sustained fashion, peripheral oscillators have been thought to damp rapidly when disconnected from the control exerted by the SCN. Using two reporter s...
متن کاملFeeding and circadian clocks.
The mammalian genome encodes at least a dozen of genes directly involved in the regulation of the feedback loops constituting the circadian clock. The circadian system is built up on a multitude of oscillators organized according to a hierarchical model in which neurons of the suprachiasmatic nuclei of the hypothalamus may drive the central circadian clock and all the other somatic cells may po...
متن کاملCircadian Clocks in Antennal Neurons Are Necessary and Sufficient for Olfaction Rhythms in Drosophila
BACKGROUND The Drosophila circadian clock is controlled by interlocked transcriptional feedback loops that operate in many neuronal and nonneuronal tissues. These clocks are roughly divided into a central clock, which resides in the brain and is known to control rhythms in locomotor activity, and peripheral clocks, which comprise all other clock tissues and are thought to control other rhythmic...
متن کاملNPAS2 Compensates for Loss of CLOCK in Peripheral Circadian Oscillators.
Heterodimers of CLOCK and BMAL1 are the major transcriptional activators of the mammalian circadian clock. Because the paralog NPAS2 can substitute for CLOCK in the suprachiasmatic nucleus (SCN), the master circadian pacemaker, CLOCK-deficient mice maintain circadian rhythms in behavior and in tissues in vivo. However, when isolated from the SCN, CLOCK-deficient peripheral tissues are reportedl...
متن کاملCentral and peripheral clocks are coupled by a neuropeptide pathway in Drosophila
Animal circadian clocks consist of central and peripheral pacemakers, which are coordinated to produce daily rhythms in physiology and behaviour. Despite its importance for optimal performance and health, the mechanism of clock coordination is poorly understood. Here we dissect the pathway through which the circadian clock of Drosophila imposes daily rhythmicity to the pattern of adult emergenc...
